The Department of Immunology, Genetics and Pathology at Uppsala University has a broad research profile with strong research groups focusing on, among other things, cancer, genetic and autoimmune diseases. A key driving force at the department is to stimulate translational research and thereby promote closer collaboration between medical research and healthcare. Research is conducted within six research programs: cancer precision medicine, cancer immunotherapy, genomics and neurobiology, molecular tools and functional genomics, neuro-oncology and neurodegeneration, and vascular biology. Parts of the operations are also integrated with the departments of oncology, clinical genetics, clinical immunology, clinical pathology, and hospital physics at Uppsala University Hospital. The department has teaching responsibilities for several program courses, standalone courses, and international master's programs within the Faculty of Medicine. The department has a turnover of approximately 550 million SEK, of which about two-thirds are external research grants. The number of employees is approximately 400, of which about 100 are doctoral students, and there are a total of over 850 active staff members. Job Description

This position is within the field of lymphatic vessel development, and the successful candidate will be responsible for conducting scientific projects, including conceptualization, planning, execution, troubleshooting, and completion via manuscript preparation.

The work will focus on:

  • Zebrafish-specific techniques including:
  • Animal husbandry and maintenance of lines.
  • Generation of mutant lines using CRISPR technology, generation of reporter lines using BAC and knock-in methods.
  • Embryo microinjection, larval angiography, and probe feeding.
  • Whole-mount immunostaining and in situ hybridization.
  • Vascular phenotypic characterization of mutant phenotypes.
  • Cell tracing using multispectral staining systems.
  • Clearing and immunostaining of juvenile samples.
  • Ex vivo imaging of adult tissues.
  • Molecular biology techniques: DNA isolation, PCR, gel electrophoresis, RNA extraction, cDNA synthesis, RT-PCR analysis, protein isolation, Western Blot, and ELISA.
  • Cell culture: stem cell differentiation into endothelial cells, culture of primary cell lines: HUVEC, HDLEC, and HDBEC, culture of tumor cell lines. Plasmid transfection, siRNA knockdown.
  • Flow cytometry: cell dissociation, staining and analysis, cell sorting in tubes and plates.
  • Imaging skills: Brightfield, Fluorescence, Confocal, Multi-photon (2-photon and 3-photon), Stellaris, Light-sheet microscopy, and Automated systems (Acquifer).
  • Data imaging analysis: ImageJ, Arivis, and IMARIS.
  • RNA sequencing: preparation of bulk-RNA libraries for MARSseq and SMARTseq2, preliminary data analysis. Collection of single-cell data and collaboration with bioinformaticians for analyses.
  • Data analysis: Prism, GraphPad for statistical analysis.
  • Teaching: supervision of students in lab projects and knowledge sharing with junior team members.
  • Science communication: poster and oral presentations at international conferences, lab meetings, and department seminars.
  • Scientific writing, peer review, and interaction with editors.
  • Organizational skills: project management, grant writing, and organization of seminars/workshops.

Qualifications

  • PhD in developmental biology, biotechnology, or biomedical sciences.
  • Experience in developmental biology and lymphatic development in zebrafish.
  • Holder of a FELASA Category B animal license or equivalent license to work with animals in Sweden.
  • Personal soft skills: Motivated and enthusiastic, cooperative, detail-oriented, good communication skills.

Desirable and/or additional merits

  • 10+ years of experience with zebrafish.
  • Multi-photon and Light-sheet imaging.
  • Experience with tissue clearing methods.
  • Project management competence.
  • Experience in supervising students.

About the Position
